Will the iPhone 16 be available to pick up in store tomorrow if upgrading?

Unless you pre-ordered it then the chances are slim to nil as pre-orders have been pushed to 3-4 weeks for delivery

Best thing to do is pop into Store and ask, but expect disappointment

You would have to ask the store concerned on that.

If you have pre ordered you should get an email. If not it's pot luck

This is not O2 and we are all customers here similar to yourself and cannot answer account type queries.
