I called up on Monday and had my number transferred from another SIM card to the SIM card that currently in my new phone. How long does this take ? And will this change on the “my number” section and on the my 02 app as well?

Your MyO2 will automatically change when the port takes place.

Did you get a text telling you when it will change?

If not, call again or complete online request https://www.o2.co.uk/help/phones-sims-and-devices/how-to-keep-your-mobile-number

Usually actioned the next day after request and changes the day after, so 48 hours, except the weekends when nothing happens.

Yes, it should be but it may not have registered on the system if you didn't get a confirmation text.
